Background
FOXG1 (Forkhead box G1) is a winged-helix transcription factor essential for vertebrate forebrain development, acting as a pioneer factor that opens chromatin and orchestrates the earliest steps of telencephalon specification by repressing dorsalizing genes, promoting FGF8 expression, and directing the temporal progression of cortical progenitor proliferation, neuronal layer fate transitions, oligodendrocyte maturation and postnatal myelination through dosage-sensitive interactions with morphogen signals, histone-modifying complexes and post-translational phosphorylation that control its nuclear–cytoplasmic shuttling, so that even modest alterations in FOXG1 levels produce a spectrum of neurodevelopmental disorders ranging from microcephaly and corpus callosum agenesis to severe encephalopathy with autism-like behaviors.
