Background
The norepinephrine transporter (NET), a 617-residue member of the Na⁺/Cl⁻-dependent neurotransmitter transporter family, is an α-helix-rich plasma-membrane protein that clears released norepinephrine from the synaptic cleft back into presynaptic noradrenergic terminals by coupling NE uptake to the co-transport of Na⁺ and Cl⁻ and the counter-transport of K⁺, thereby terminating noradrenergic signaling, regulating sympathetic and central adrenergic neurotransmission, controlling heart rate and blood pressure, and serving as the molecular target for drugs such as atomoxetine, reboxetine and amphetamines; NET dysfunction is implicated in ADHD, depression, orthostatic intolerance and neurodegenerative diseases, and its availability at the cell surface is dynamically modulated by kinase phosphorylation, protein–protein interactions with syntaxin 1A and PICK1, and trafficking between endosomal compartments, making it a key determinant of noradrenergic homeostasis and a focus of neuropsychiatric therapeutic strategies.
